5.2.7 Challenge testing during process
validation should be avoided as product
development and any pre-validation/engineering/trial runs should have already
defined the process parameters and identified any causes of variation. Challenge tests (only within the established parameters)
might however be useful to close any remaining minor gaps arising from the risk
assessment. There is also the possibility to perform additional
testing during the subsequent ongoing process verification to address
any issues so long as this is documented and
justified.
5.2.7 工艺验证期间应避免挑战性测试,因为产品开发以及任何预验证/工程/试运行应已定义工艺参数并识别出任何变异原因。然而,挑战性测试(仅在既定参数范围内)可能有助于弥补风险评估中产生的任何剩余微小差距。在后续持续工艺核实期间进行额外检测以解决任何问题也是可能的,前提是这已记录并论证。
二:说下之前结合实际经验开展的情况:
比如固体制剂的压片,都有一个参数范围,当然这个参数范围是之前早期的工艺设计已经被验证通过的合理参数范围(不包括非合法,或者挑战的极限),实际运营过程中,会有“高、中、低”三类参数。验证中,是执行了2+1+1,即2批中间压片速度,1批高的速度,1批低的速度,审计过程中,无异议。这种不是挑战测试,而是符合PICS的2026版确认与验证指南提到的在既定参数范围内的挑战测试,而且是被记录论证合理过。
